Glowing Bacteria Lab Essay 1. Transformation involves the transfer of genetic information into a cell by directly taking up foreign DNA from its surroundings. This DNA is then incorporated into the host cell’s own DNA. This transformation usually occurs within plasmids, small circular DNA molecules separate from its chromosome. After the recipient cell is infected with the DNA, the cell will move on with replication, producing offspring with traits encoded by the plasmid. These plasmids may replicate with the chromosome, or independently. This is how diseases are commonly spread, since one little bit of DNA can affect the entire organism thanks to duplication. 2. E. Coli are ideal organisms for molecular geneticists to manipulate because it can easily be grown in suspension culture in mediums such as Luria broth or on agar. Also, E. Coli has a relatively small genome, containing only about five million DNA base pairs. By chemically and thermally treating E. coli cells, they can artificially be transformed. Naturally, these cells do not possess the natural system needed for transformation. After treatment, they become receptive to an insertion of foreign DNA contained in a plasmid. 3. After treating the E. coli cells, plasmid containing firefly genes can be inserted into the cell. What this will do is create recombinants in which properties of the original cell and the firefly cell will be exhibited. In order to insert the plasmid into the cells, we will use a sterile inoculating loop to remove a loopful of the E. Coli from the surface of the agar. We will then place the loop into calcium chloride and twirl it rapidly, releasing the bacteria. Then, using a pipette, we will add 10 µl of the pBestLuc solution to the mixture. 4. Luciferase is the enzyme responsible for the luminescent glow of the firefly. It causes the glow by catalyzing a reaction between the chemicals luciferin and ATP in the presence of oxygen and magnesium. After injecting the cells with this enzyme, and allowing them to sit for 24 hours, we will be able to observe whether or not transformation was successful based on whether or not our bacteria will glow. 5. The way in which we will calculate the efficiency of the transformation reaction is by comparing our results with those of our classmates. If most of the groups did not have successful transformation, it is safe to conclude that the efficiency is relatively low. On the otherhand,  if all the groups see that the bacteria has taken on the new trait, than the efficiency is relatively high.

How The Adams-Onà ­s Treaty Affected The Growth Of Our Nation Americans were interested in further expansion and looked to the weak Spanish provinces of East and West Florida. The Spanish were reluctant to give up what is now Florida, but in the end they worked out an agreement called the Adams Onis Treaty. In this essay I will describe how the Americans eventually got these provinces, the set backs of the signing of the treaty, and how it effected the economic growth of our nation. Americans living in West Florida between the Iberville and Perdido Rivers declared their independence, and President Madison ordered the Governor of New Orleans Territory to take control of the independent land. The Americans now had control over half the territory they wanted but this did not satisfy them, this only made them more eager to gain control of East Florida. Almost two years after this event peace concluded, but Spain still had possession of East Florida. This time Spain only had two solutions to avoid a shameful political break down over this region. One was to gain support of a European ally, and the second was to get some form of honor in this event by winning some of the United States best land else where in North America. The first plan fell through, Britain and other European nations refused to help Spain. They eventually fell to having discussions with the United States. The American government was entirely willing to have discussion of the East Florida issue become the end result of all Spanish-American boundary questions. John Quincy Adams was the Secretary of State and was the person taking care of all discussions between the U.S. and Spain. He saw that this was a perfect time to try to extend the boundary to the Pacific Ocean. Luis de Onà ­s was the minister of Spain at the time and he was the Spanish representative to the U.S. His instructions from the Spanish government was to transfer the Florida's to the United States in return for the American settlement west of the Mississippi. Onis was also to get a promise that the United States would not give material aid to, or recognize the independence of Spain's colonies in South America because of their lack of cooperation. Difficulties came up over negotiating a satisfactory boundary in the West and also over the command to put American military in East Florida in 1818.

Illinois case allowed states to regulate certain businesses within their borders, including railroads. The Great Railroad Strike of 1877 began on July 14 in Martinsburg, West Virginia, United States and ended some 45 days later after it was put down by local and state militias, as well as by federal troops. Hayes essentially sold out his ideals to the former Confederacy in order to gain the Presidency. He allowed the former Confederate states to return to governing themselves almost the same exact way they had been doing prior to the Civil War. This essentially set the Civil Rights movement back for almost a century, as many â€Å"Jim Crow† Laws were passed to ensure that blacks and other minorities could not rise to the same equality as whites, laws which were enforced in the Supreme Court case of Plessy v. Fergusson, which established the â€Å"separate but equal† mantra, and later overturned in the case of Brown v. the Board of Education of Topeka, Kansas. As the public†™s view of nursing and nurses becomes skewed, behavior that was previously seen as wrong could now be seen somewhat acceptable.
